Reduced forms of nitrogen are required for the biosynthesis of amino acids and nucleic acids, and the only enzyme capable of this complex, multi-electron reduction is nitrogenase. The catalytic mechanism of nitrogenase is highly dynamic and involves nucleotide-dependent protein-protein associations/dissociations, conformational changes in the proteins, and many oxidation and structural states of the metalloclusters. Using cryoEM, we have structurally characterized the nitrogenase complex during catalytic turnover, providing the first observations of nucleotide-dependent conformational changes around the catalytic site.
